Product Name :
Goat anti Human C1 esterase inhibitor

Description :
| Clone Polyclonal | Units 1 ml | Host Goat

Background :
C1 esterase inhibitor is a single chain acid glycoprotein with a molecular weight of 110,000. The protein has a high carbohydrate content. In immunoelectrophoresis it ha an alpha mobility. The concentration in normal serum ranges from 150 to 350 μg/ml. Reduced levels are observed in hereditary angioedema. It regulates the activation of complement factor C1 as well as the activity of activated C1 by coupling with the active catalytic site at the light chains of C1r and C1s. Moreover it also inhibits, kallikrein, Factor XI and Factor XII. Freund’s complete adjuvant is used in the first step of the immunization procedure.

Source :
The reactivity of the antiserum is restricted to C1 esterase inhibitor. In immunoelectrophoresis and radial immunodiffusion (Ouchterlony), using various antiserum concentrations against fresh normal human plasma a single precipitin line is obtained which shows a reaction of identity with the precipitin line obtained with purified C1 esterase inhibitor. No reaction is obtained with any other plasma protein component or serum. However, the antiserum may also react with fibrin monomers, circulating fibrinopeptides and fibrin degradation products. In precipitating techniques as immunoelectrophoresis and single or double radial immunodiffusion to identify the presence of C1 esterase inhibitor in human plasma or other body fluids or to determine its concentration. The presence of non-precipitating antibodies has not been assayed. This does not exclude the use of the antiserum in non-precipitating antibody-binding techniques if proper controls are included. In immunoelectrophoresis use 2 μl or equivalent against 120 μl antiserum. In double radial immunodiffusion (Ouchterlony) use a rosette arrangement with 10 μl in a 3 mm diameter center well and 2 μl serum samples (neat and serially diluted) in 2 mm diameter peripheral wells. <

Product :
Delipidated, heat inactivated, lyophilized, stable whole antiserum No preservative added Total protein and IgG concentrations in the antiserum are comparable to those of pooled normal goat serum. No foreign proteins added. Reconstitute the lyophilized antiserum by adding 1 ml sterile distilled water. Formulation: Delipidated, heat inactivated, lyophilized, stable whole antiserum No preservative added Total protein and IgG concentrations in the antiserum are comparable to those of pooled normal goat serum. No foreign proteins added.

Specificity :
Precipitating polyclonal Goat antiSerum to Human C1 esterase inhibitor Species Reactivity: The antiSerum does not cross react with any other component of Human plasma. Inter-species cross-reactivity is a normal feature of antibodies to plasma proteins since they frequently share antigenic determinants. Cross-reactivity of this antiSerum has not been tested in detail.

Applications :
Precipitation assays. Dilutions may be prepared by adding phosphate buffered saline (PBS, pH 7.2). Repeated thawing and freezing should be avoided. If a slight precipitation occurs upon storage, this should be removed by centrifugation. It will not affect the performance of the antiserum. Diluted antiserum should be stored at +4°C, not refrozen, and preferably used the same day.

Storage :
The lyophilized antiserum is shipped at ambient temperature and may be stored at +4°C; prolonged storage at or below -20°C.

Antibodies are immunoglobulins secreted by effector lymphoid B cells into the bloodstream. Antibodies consist of two light peptide chains and two heavy peptide chains that are linked to each other by disulfide bonds to form a “Y” shaped structure. Both tips of the “Y” structure contain binding sites for a specific antigen. Antibodies are commonly used in medical research, pharmacological research, laboratory research, and health and epidemiological research. They play an important role in hot research areas such as targeted drug development, in vitro diagnostic assays, characterization of signaling pathways, detection of protein expression levels, and identification of candidate biomarkers.
